Company overview
Phase 3 Neurodegenerative Disease Company
Annovis Bio is developing buntanetap, an oral candidate for Alzheimer’s disease and Parkinson’s disease. Through an RNA-targeting mechanism, buntanetap is designed to reduce translation of multiple neurotoxic proteins, including APP/amyloid beta, tau, alpha-synuclein, and TDP-43.
Registrational Alzheimer’s Study
The pivotal Phase 3 Alzheimer’s study enrolls patients with early disease and a positive plasma pTau217 biomarker. The design evaluates symptomatic improvement at six months and a potential disease-modifying effect over 18 months.
Biomarker-Enabled Development
Annovis is incorporating plasma pTau217, skin-biopsy alpha-synuclein, and digital motor assessments across its programs. These tools are intended to enrich patient selection and connect clinical outcomes with evidence of underlying disease biology.
One Oral Asset Across Two Diseases
Buntanetap is being developed as a once-daily oral therapy across both Alzheimer’s and Parkinson’s disease. A shared mechanism and manufacturing base allow Annovis to pursue two large neurodegenerative indications while using disease-specific clinical endpoints and biomarkers.
Long-Term Parkinson’s Study
An open-label extension study in Parkinson’s disease is assessing safety and efficacy over 36 months. It incorporates skin-biopsy alpha-synuclein biomarkers and NeuroRPM artificial intelligence-based digital measures to track changes in motor function.
Future catalysts
Randomization in the Alzheimer’s Phase 3 study is expected to be completed in summer 2026, with initial topline results anticipated in early 2027.
An NDA for buntanetap as a symptomatic Alzheimer’s treatment is expected in early 2027, followed by a potential disease-modifying treatment submission in early 2028.
Enrollment in the Parkinson’s open-label extension study is expected to be completed in the fourth quarter of 2026.
