ARCUS Gene Editing Platform

Precision BioSciences develops in vivo gene-editing therapies using ARCUS nucleases. The compact enzymes are designed for several editing modes, including elimination of viral DNA, excision of defective genomic regions and targeted insertion of functional genetic material.

PBGENE-HBV

PBGENE-HBV is a wholly owned in vivo editing therapy in the Phase 1/2a ELIMINATE-B trial for chronic hepatitis B. It is designed to eliminate covalently closed circular DNA, the source of infectious viral replication, while inactivating integrated HBV DNA.

PBGENE-DMD

PBGENE-DMD uses two ARCUS nucleases delivered in one AAV to excise exons 45 through 55 of the dystrophin gene. The approach is intended to restore a near-full-length functional dystrophin protein in a large subset of patients with Duchenne muscular dystrophy and is entering the Phase 1/2 FUNCTION-DMD trial.

Partnered In Vivo Programs

iECURE is developing ECUR-506, an ARCUS-mediated gene-insertion therapy for neonatal-onset ornithine transcarbamylase deficiency. The OTC-HOPE study is evaluating whether targeted insertion can produce durable metabolic correction.

Partnered Cell Therapy

The allogeneic CAR-T candidate azer-cel is licensed to partners for development in lymphoma, progressive multiple sclerosis and other autoimmune diseases. These programs provide external validation and potential economics from an earlier ex vivo application of ARCUS technology.
