Long-Acting Immunology Antibodies

SKYLINE IBD Platform

The Phase 2 SKYLINE platform trial evaluates SPY001, SPY002 and SPY003 as monotherapies and in pairwise combinations in ulcerative colitis. The antibodies target α4β7, TL1A and IL-23, respectively, enabling Spyre to compare individual mechanisms and combination effects within one development framework.

SPY001

SPY001 is a high-concentration, half-life-extended α4β7 antibody designed for infrequent maintenance dosing. Initial induction data from SKYLINE supported continued development and provided the first clinical proof of concept for Spyre’s platform.

SKYWAY Rheumatic-Disease Program

The Phase 2 SKYWAY basket trial evaluates the anti-TL1A program SPY072 in rheumatoid arthritis, psoriatic arthritis and axial spondyloarthritis. This trial tests whether the same long-acting immunology approach can extend beyond IBD into multiple rheumatic diseases.
